Pyridoxal phosphate (PLP, the main metabolically active form of vitamin B6) is a multifunctional coenzyme that participates in the metabolism of amino compounds. Enzymes that use it as a coenzyme factor are called PLP-dependent enzymes (PLP-DEs). They play an important role in the metabolism of sugars, lipids, and amino acids, mainly in the biosynthesis of amino acids and biomolecules, such as neurotransmitters serotonin, dopamine, adrenaline, norepinephrine, γ-aminobutyric acid (GABA), and histamine.

PLP is so important that it is involved in more than 140 PLP-dependent enzyme activities, accounting for about 4% of all classified activities. The versatility of PLP comes from its electrophilicity: PLP covalently binds to the substrate by forming an external aldimine at the beginning of the reaction, and then different types of carbon anion reaction intermediates can be stabilized by PLP through resonance effects. This function of PLP is also called "electron trap". In addition to the electron absorption effect, since almost all PLP-dependent reactions generate cationic and anionic intermediates, acidic and basic groups that assist proton transfer are equally important.
